sAC C2 KO Mouse

Description: Soluble adenylyl cyclase (sAC; Adcy10) C2 knockout mice were generated by deleting a 2.2 kb genomic region encompassing exons 11–13, which encode a substantial portion of the C2 catalytic domain. The targeting construct incorporated flanking loxP sites and an FRT-flanked neomycin selection cassette, enabling removal of the selection marker via Flp recombinase and subsequent Cre-mediated excision. Following homologous recombination in 129Sv embryonic stem cells and blastocyst injection, mice were bred to Flp-expressing strains to generate floxed (Adcy10lox) alleles, and then to Cre-expressing strains to produce germline deletion (C2KO) mice lacking the catalytic C2 domain of sAC.

Applications: Mouse model for studying the role of soluble adenylyl cyclase in cAMP signaling, including its functions in metabolism, neuronal signaling, and fertility.
